PUB HLTH 7119 MPH Dissertation Question: A modified Folin–Ciocalteau method was carried out to mea-sure free and bound phenolics of the extracts (Fang, Zhang, Sun,& Sun, 2006), using gallic acid as a standard (0–360 mg/l). The freeor bound phenolic extracts (100 ll) were mixed with 2.5 ml of0.2 N Folin–Ciocalteau reagents, and then 2 ml of saturated sodiumcarbonate solution (75 g/l) were added. The absorbance of the mix-ture was determined at 765 nm using an UV-1800 Spectropho-tometer (Shimadzu, Canby, USA) after reaction for 2 h at roomtemperature in the dark. Results were expressed as mg gallic acidequivalents (GAE)/g dry basis, (db). Total phenolic content was calculated as the sum of free and bound.  Answer: A Folin-Ciocalteau which is a modified method was employed to take the measurement of bound and free phenolics of the excerpts. This was carried by the use of gallic acid as a standard of 0-360mg/l. Folin-Ciocalteau of 2.5 ml of 0.2 N was mixed with extracts of bound or free phenolic reagents. After that about 2ml of sodium carbonate solution which is saturated and of the amount of 7.5 g/l were added. The mixture´s absorbance was obtained at 765nm by the means of UV-1800 spectrometer (Peter, 2013). This reaction was conducted for a period of two hours in a dark room and at room temperature. The results from the experiment were put as milligram gallic acid equivalent per dry basis db. The sum of phenolic content was obtained as the total of the bound and free.
